Telomere dysfunction drives increased mutation by error-prone polymerases Rev1 and zeta in Saccharomyces cerevisiae.

Using a model system, we have shown that replicative senescence is accompanied by a 16-fold increase in base substitution and frameshift mutations near a chromosome end. The increase was dependent on error-prone polymerases required for the mutagenic response to DNA lesions that block the replication fork.

Error-prone repair DNA polymerases in prokaryotes and eukaryotes.

DNA repair is crucial to the well-being of all organisms from unicellular life forms to humans. A rich tapestry of mechanistic studies on DNA repair has emerged thanks to the recent discovery of Y-family DNA polymerases. Many Y-family members carry out aberrant DNA synthesis-poor replication accuracy, the favored formation of non-Watson-Crick base pairs, efficient mismatch extension, and most i...

Error-Prone Candidates Vie for Somatic Mutation

Antibodies play a major role in the resistance of higher organisms to disease. This is made possible because mice, humans, and other species have evolved three unusual molecular mechanisms that allow the generation of an enormously diverse repertoire of antibodies from a relatively small amount of genetic material. First, each antibody-forming B cell assembles different combinations of variable...

Estimation of the Mutation Rate During Error-prone Polymerase Chain Reaction

Error-prone polymerase chain reaction (PCR) is widely used to introduce point mutations during in vitro evolution experiments. Accurate estimation of the mutation rate during error-prone PCR is important in studying the diversity of error-prone PCR product. Although many methods for estimating the mutation rate during PCR are available, all the existing methods depend on the assumption that the...

Expression of error-prone polymerases in BL2 cells activated for Ig somatic hypermutation.

High affinity antibodies are generated in mice and humans by means of somatic hypermutation (SHM) of variable (V) regions of Ig genes. Mutations with rates of 10(-5)--10(-3) per base pair per generation, about 10(6)-fold above normal, are targeted primarily at V-region hot spots by unknown mechanisms. We have measured mRNA expression of DNA polymerases iota, eta, and zeta by using cultured Burk...
